葛宏华博士，1973年出生，安徽巢湖人，安徽大学健康科学研究院 教授级高级工程师，硕士生导师。先后毕业于安徽大学生物化学专业（获理学学士学位）和中国科学技术大学生化与分子生物学专业（获理学博士学位）。2008年至2009年在澳大利亚Monash 大学Jamie Rossjohn 实验室从事博士后研究工作。目前的研究领域为蛋白质的结构与功能，主要针对嗜肺军团菌毒力因子蛋白以及其他重要蛋白质进行结构生物学研究。主持的在研项目包括国家自然科学基金面上项目（2013-2016）、安徽省自然科学基金项目（2011-2013）以及安徽省高校省级自然科学研究重点项目（2011-2013）等。

1. Legionella pneumophila, the causative agent of Legionnaires' disease, is a Gram-negative facultative intracellular pathogen capable of multiplying in a wide spectrum of eukaryotic cells. The pathogenicity of L.pneumophila is entirely dependent upon its ability to replicate within host cell, a process that requires the Dot/Icm type IV secretion system (T4SS). The Dot/Icm transporter injects a large number of virulence factors into host cells and many of these virulence factors benefit the bacterium by controlling host membrane transport.
Our research will focus on the structural and functional characterization of some factors. Functional and crystallographic studies will provide fundamental insights into the structural basis as well as the molecular mechanism of these factors in host cells. The structural and functional information will lay the foundation for the development of novel prevention and therapeutic strategies against diseases caused by L. pneumophila and possibly other infectious agents. It will also provide new tools and reagents invaluable in the investigation of host processes.
2. Laccases are oxidoreductases that belong to the multinuclear copper-containing oxidases. They are able to oxidize a wide range of substrates, preferably phenolic compounds, which makes them suitable for employment in the bioremediation of soil and water as well as in other biotechnological applications. We use structure approaches, as well as biochemical technique to address the details of laccase oxidation mechanism.
